One-tube oxidation-fermentation methods: limitations posed by atypical fermentative reactions.
نویسنده
چکیده
Atypical fermentation in the two-tube method of Hugh and Leifson (1953), whereby acid was produced in the sealed tube but not in the open tube, was determined to be the result of a preferential metabolism of peptone over the reactant carbohydrate. These atypical fermentations are only partially detected by one-tube oxidation-fermentation methods, and as such they constitute a source of limitation to the routine use of these abbreviated techniques for oxidative-fermentation determinations.

One-tube test for determining oxidative or fermentative metabolism of bacteria.
A one-tube test for distinguishing oxidation from fermentation in the metabolism of carbohydrates was examined on 1,200 gram-negative bacilli. There was a 100% correlation between the one-tube method and the standard two-tube procedure.
متن کاملModified medium for the oxidation-fermentation test in the identification of marine bacteria.
Using the medium of ZoBell as a basal formulation, we developed a medium to determine the oxidative-fermentative metabolism in marine bacteria. Different oxidative and fermentative collection strains, as well as a group of unidentified marine bacteria, were tested on modified ZoBell medium and other conventional media. In general, superior results were obtained with the modified ZoBell medium s...
متن کاملA Study of Two Atypical Strains of E. typhosa.
In 1937, two atypical strains of Eberthella typhosa were isolated twenty-five days apart from patients presenting clinical symptoms of typhoid fever. Blood and feces from one patient, and blood from the other, yielded organisms atypical of E. typhosa in that they showed delayed fermentation of lactose and sucrose, but failed to produce hydrogen sulfide. Both patients gave a history of having ea...
متن کاملMetabolically engineered bacteria for producing hydrogen via fermentation
Hydrogen, the most abundant and lightest element in the universe, has much potential as a future energy source. Hydrogenases catalyse one of the simplest chemical reactions, 2H(+) + 2e(-) ↔ H(2), yet their structure is very complex. Biologically, hydrogen can be produced via photosynthetic or fermentative routes. This review provides an overview of microbial production of hydrogen by fermentati...
متن کاملCoupling of Reduced Pyridine Nucleotide in Leuconostoc Mesenteroides.
The demonstration that oxidation-reduction reactions mediated by the pyridine nucleotide coenzymes occur with the transfer of hydrogen between substrate and coenzyme (1) has led to the use of hydrogen isotopes to measure the occurrence of the specific coupling of reactions in the intact cell and organism (2-4). Several problems associated with the cellular organization and integration of the fe...
